Biological Activity
ACF-01 is a small molecule activator of prolyl hydroxylase domain protein 2 (PHD2), directly binds and activates PHD2 (SPR KD=41.7 uM), thereby promoting HIF-1α degradation.
ACF-01 promotes PHD2-mediated hydroxylation, leading to reduced stability and accelerated pVHL-dependent degradation of HIF-1α.
ACF-01 synergizes with osimertinib to downregulate HIF-1α expression on NSCLC cells.
Combination of ACF-01 and osimertinib impairs HIF-1α–driven glycolysis and angiogenesis, enhances the antiproliferative and pro-apoptotic effects of osimertinib.
ACF-01 improves the antitumor efficacy of osimertinib in NSCLC xenograft model.
