PIP4K-IN-a131 (PIP4K inhibitor a131) is a PIP4K lipid kinase inhibitor with IC50 of 1.9 uM (PI5P4Kα), selectively kills cancer cells, while protecting normal cells.
PIP4K-IN-a131 inhibits the kinase activity of purified PIP4K2A in vitro as well as PIP4Ks activity with IC50 of 1.9 uM and 0.6 uM, respectively.
Ras activation overrides a131-induced PIK3IP1 upregulation and activates the PI3K/Akt/mTOR pathway.
