PRC1590 is a tetrahydro-β-carboline compound that potently kills the malaria parasite with EC50 of 145 nM and 142 nM in growth inhibition assays in 3D7WT and Dd2 strains.
PRC1590 has a mechanism of action associated with P. falciparum's DV that differs from chloroquine, inhibits trophozoite stage.
The mechanism of resistance to PRC1590 is mediated by the G293V single-nucleotide polymorphism (SNP) of the P. falciparum multidrug resistance protein 1 (PfMDR1), a transporter located on the membrane of the digestive vacuole (DV).
