PST inhibitor peptide-8 (PSTi8) is a potent pancreastatin (PST) inhibitor, competes with PST on GRP78 receptor binding and rescues PST-induced insulin resistance (IR) both in vitro and in vivo.
PST (100 nM) was found to inhibit >50% binding of labelled PSTi8 (150 nM) and PSTi8 (300 nM) was found to inhibit >50% binding of labelled PST (25 nM).
PST (100 nM) inhibits protein expression of GRP78 in tunicamycin stimulated condition in HepG2 cells, which is significantly inhibited by PSTi8 (800 nM), PSTi8 also enhances tunicamycin effect on stimulation of GRP78 protein expression.
PSTi8 regulates secreation of biomolecule, glycogen storage and hepatic gene expression.
PSTi8 acute (i.p, 5 mg/kg) and chronic (i.p, 2 mg/kg for 7 days), administration attenuates IR in HFD-fed, HFrD-fed and db/db diabetic mice.
