PTP1B inhibitor BDB is a natural compound found in marine red algaea, is a selective, cell‐permeable and competitive inhibitor of PTP1B with IC50 of 1.86 uM.
PTP1B inhibitor BDB binds to PTP1B with an affinity of 1.812 μM and in a slow‐on and slow‐off manner in SPR assays.
PTP1B inhibitor BDB enhances the stability of PTP1B, protects PTP1B from pronase‐induced proteolysis.
PTP1B inhibitor BDB is selective for PTP1B over other PTPs (TC‐PTP, SHP‐1, SHP‐2, and LAR).
PTP1B inhibitor BDB (50 and 100 mg/kg/day, oral, 9 weeks) protects PTP1B protein from temperature‐dependent degradation at the intact‐cell level.
PTP1B inhibitor BDB prevents hyperglycaemia in diabetic BKS db mice, improves glucose clearance and significantly decreases insulin resistance in diabetic mice.
