Biological Activity

PTPN22 inhibitor 8b-19 is a potent, selective inhibitor of protein tyrosine phosphatase non-receptor type 22 (PTPN22) with IC50 of 0.25 uM, 15.2-fold and 19.2-fold selectivity against SHP2 and PTP1B, respectively.
PTPN22 inhibitor 8b-19 displays little to no activity against an extended panel of mammalian PTPs including nonreceptor PTPs, SHP1, TCPTP, MEG2, PEST, HePTP, and FAP-1, receptor-like PTPs, PTPα and PTPε, dual specificity PTPs, VHZ and Laforin, and the low molecular weight PTP.
PTPN22 inhibitor 8b-19 (10 uM) increases T cell signaling in a dose-response manner, induces greater levels of phosphorylated LCK and ERK1/2 in Jurkat cells.
