PTPN22 inhibitor NC1 is a specific, noncompetitive, allosteric PTPN22 (lymphoid-specific tyrosine phosphatase, LYP) inhibitor with Ki of 4.3 uM, effectively and specifically inhibits LYP-mediated TCR signaling in T cells.
PTPN22 inhibitor NC1 displays at least 1.9-fold higher selectivity against a panel of other protein phosphatases, including STEP, PTPN18, Glepp, VHR, and the Ser/Thr phosphatases PPM1A and PP1.
PTPN22 inhibitor NC1 (20 uM) significantly increased the phosphorylation levels of ERK and LCK in Jurkat T cells, shows no effect on CD3-induced LCK and ERK phosphorylation in T cells without endogenous LYP expression.
