PW0729 is a potent, selective, G protein-biased agonist of orphan GPR52 with EC50 of 47 nM in cAMP assay, Emax=143%.
PW0729 shows EC50 of 110 nM in β-arrestin recruitment assay.
PW0729 is highly selective for GPR52 agonism with no significant off-target effects at other brain GPCRs.
PW0729 is also bioavailable but hs less than optimal brain permeability, requiring further optimization.
