Pyr3 is a potent, selective inhibitor of transient receptor potential canonical channel 3 (TRPC3) with IC50 of 700 nM for inhibition of TRPC3-mediated Ca2+ influx.
Pyr3 shows no effect on Ca2+ influx mediated by other TRPC members, Ca2+ responses in the absence of extracellular Ca2+, and the control basal [Ca2+]i levels.
Pyr3 potently eliminates the Ca2+ influx-dependent PLC translocation to the plasma membrane and late oscillatory phase of B cell receptor-induced Ca2+ response in DT40 B lymphocytes.
Pyr3 attenuates activation of nuclear factor of activated T cells, a Ca2+-dependent transcription factor, and hypertrophic growth in rat neonatal cardiomyocytes, and in vivo pressure overload-induced cardiac hypertrophy in mice.
