RO 5263397 is a highly potent, selective and orally bioavailable trace amine-associated receptor 1 (TAAR1) partial agonist with EC50 of 17 nM, >1,000-fold selectivity over human adrenergic α2A receptor.
RO 5263397 shows EC50 of 35 nM for rat TRRA1.
RO 5263397 blocks psychostimulant-induced hyperactivity and produces a brain activation pattern reminiscent of the antipsychotic drug olanzapine in vivo, does not induce catalepsy or weight gain, promotes vigilance in rats and shows pro-cognitive and antidepressant-like properties in rodent and primate models.
