Biological Activity

RO8323 is a potent and selective dual CDK8/19 inhibitor with IC50 of 2/3 nM respectively, has 100-fold kinome selectivity.
RO8323 promotes Treg differentiation and suppresses effector T cells in vitro and in vivo.
RO8323 transcriptionally represses pro-inflammatory programs in activated CD4+ T cells.
RO8323 potentiates IL-10 production in TLR-stimulated immune cells in vitro and in vivo.
RO8323 attenuates transplant rejection, prevents cGVHD and attenuates renal pathology in mice.
RO8323 increases Treg populations and enhances immune reconstitution in cGVHD mice. prolongs cardiac allograft survival in mice.
