Biological Activity
RTY-406 is a dual-acting ABCB4/BSEP (ATP-binding cassette subfamily B member 4 / bile salt export pump) positive functional modulator (PFM), increases ABCB4 and BSEP protein levels and functional output, thereby increasing biliary phospholipid secretion and bile acid efflux.
RTY-406 increased biliary phospholipids and reduced markers of cholestasis, cholangitis, ductular reaction, and fibrosis in mouse model of PSC-like cholestatic disease.
RTY-406 produced dose-dependent reductions in GGT (gamma-glutamyl transferase) and serum cholesterol, along with changes consistent with increased ABCB4 and BSEP activity in healthy cynomolgus monkeys.
