Biological Activity
RVJB59 is a covalent small molecule activator of wtP53 and p53 mutants, shows selective covalent binding to Cys 141 of wt p53DBD.
RVJB59 thermally stabilizes the R280K mutant p53 in a dose-dependent manner in differential scanning fluorimetry assays.
RVJB59 enhances the thermostability of the wt and R273H p53 DNA-binding domain.
RVJB59 is selective for cancer cells and may reactivate p53 in mutant p53 cells in cancer cell line expressing R273H mutant p53 (HT29) and in fibroblasts.
RVJB59 reduced 3D tumor spheroid growth and showed antiangiogenic potential.
