SACLAC is an α-chloroamide ceramide analog that binds irreversibly to acid ceramidase (ASAH1), significantly reduces the viability of AML cells (EC50=3 uM) across 30 human AML cell lines.
SACLAC induces apoptosis and loss of mitochondrial membrane potential, reduces cell viability and colony formation, reduces expression of SF3B1 and increases pro-apoptotic Mcl-1S.
SACLAC (5 mg/kg SACLAC in 45% w/v 2-hydroxypropyl-β-cyclodextrin in PBS) reduces leukemic burden in NSG mouse models of AML.
