Biological Activity
SAIT-PDZ-5179 is a small molecule inhibitor of GAIP-interacting protein C terminus 1 (GIPC1), selectively target the PDZ domain of GIPC1 with ITC KD of 943 nM.
SAIT-PDZ-5179 attenuates downstream signaling pathways in PDAC cells, leading to suppression of multiple downstream targets.
SAIT-PDZ-5179 induces a modest cytotoxic response in PDAC and patient-derived PDX cell lines.
SAIT-PDZ-5179 markedly reduces colony formation in PANC-1, KPC, AsPC-1, PANC-02, PDX-6741 and PDX-7114 cells.
SAIT-PDZ-5179 (10 mg/kg, twice weekly) exhibits significant antitumor activity in a syngeneic orthotopic murine model of PDAC.
SAIT-PDZ-5179 induces remodeling of immune and extracellular matrix-associated pathways in PDAC tumors.
