SAR7334 (SAR-7334 ) is a highly potent and selective inhibitor of TRPC6 channel, reduces TRPC6-mediated Ca2+ influx with IC50 of 9.5 nM.
SAR7334 also inhibits TRPC3 and TRPC7 channels at significantly higher concentrations (IC50=282 nM and 226 nM respectively), has no appreciable effects on TRPC4 or TRPC5 channels (IC50>10 uM).
SAR7334 dose-dependently reduces TRPC6 currents in patch-clamp experiments on TRPC6-HEK-FITR cells with IC50 of 7.9 nM.
SAR7334 suppresses TRPC6-dependent acute HPV in isolated perfused lungs from mice, does not change mean arterial pressure in spontaneously hypertensive rats (SHR).
