SB-328437 is a potent, selective non-peptide CC chemokine receptor-3 (CCR3) antagonist, inhibits 125I-eotaxin binding to human eosinophils with IC50 of 4.5 nM.
SB-328437 inhibits binding of 125I-MCP-4 to human eosinophils with IC50 value of 7 nM.
SB-328437 (30 uM) does not inhibit the binding of 125I-IL-8 to CHO-CXCR1 cell or CHO-CXCR2 cell membranes, 125I-SLC (CCL-21) to HEK-293-CCR7 membranes, or [3H]LTD4 to guinea pig lung membranes.
SB-328437 is 250-fold selective for CCR3 over the other 7-TM receptors tested.
SB-328437 is potent inhibitor of eotaxin- and MCP-4-induced Ca(2+) mobilization in RBL-2H3-CCR3 cells and eosinophils.
