Biological Activity
SIH-001 is a specific small-molecule inhibitor of ZMIZ1 (Zinc finger MIZ domain-containing protein 1) with SPR KD of 0.498 uM.
SIH-001 selectively reduces the intracellular stability of ZMIZ1, while exerting no detectable effects on other ZMIZ family proteins.
SIH-001 induces effective suppression of ZMIZ1 chromatin association in OCI-AML3 and MOLM-13 cells.
SIH-001 markedly potentiates AML cell differentiation and/or apoptosis induced by the menin inhibitor ziftomenib and the BCL2 inhibitor venetoclax, respectively.
SIH-001 downregulates DNA-replication and stemness-related programs and upregulated hematopoietic and antigen-presentation/surface-receptor signaling pathways.
SIH-001 significantly prolonged the survival of AML-bearing mice in both the murine AML model and AML organoids.
