Biological Activity
SIMM-036 is a specific small molecule SARS-CoV-1 PLpro inhibitor with IC50 of 192.3 nM, inhibits the K63-deubiquitinase activity of SARS-CoV-2 Plpro.
SIMM-036 does not inhibits host deubiquitinase activities (USP1, USP7 and USP28).
SIMM-036 potently blocks the PLpro mediated deubiquitinase activity against K63-linked polyUb4 chain with the IC50 value of 10.48 uM, more potent than GRL-0617.
SIMM-036 (10 uM) significantly inhibits PLpro-mediated viral replication activity, protects the RLR mediated interferon response from the K63-deubiquitinase activity of SARS-CoV-2 PLpro.
