Teplinovivint (SM04755) is a small molecule canonical Wnt signaling inhibitor, potently inhibited CLK2, CLK3, and DYRK1A with IC50 of 5.0 nM, 48.6 nM and 3.5 nM, respectively.
SM04755 inhibits inflammation and promoted in vivo tendon healing in chronic and delayed‐treatment models of tendinopathy in rats.
SM04755 dose‐dependently inhibits the CLK‐mediated phosphorylation of SRSF1, SRSF4, SRSF5, and SRSF6 in rTDSCs and hMSCs.
SM04755 induces tenocyte differentiation, reduce tendon‐destroying proteases, and decrease inflammatory cytokine production.
