SOX11 inhibitor Compound R (SOX11i) is a small molecule inhibitor of SOX11, bind directly to SOX11-DBD and interfere with DNA binding.
Compound R demosntrates SOX11-specific growth inhibition (IC50=10-12 uM) in SOX11-positive MCL cell lines (JeKo-1, Mino and Z-138).
Compound R inhibit SOX11-dependent signaling and gene expression, inhibits pBTK levels in all three SOX11+ cell lines (IC50=23-34 uM), no effect on pBTK level in SOX11-negative JVM-2 cells.
Compound R induces cell death in Ibrutinib-resistant primary MCL and is synergistic with Ibrutinib in SOX11+ MCL cell lines.
SOX11 belongs to the SOXC family of high mobility group (HMG) transcription factors, which consists of SOX4, SOX11, and SOX12. SOX11 protein is expressed in the majority (78–93%) of MCL patients and is specific for MCL as compared to other types of Non-Hodgkin's Lymphoma (NHL).
