SRC-3 inhibitor SI-12 is a potent small molecule inhibitor of steroid receptor coactivator (SRC-3), inhibits MCF-7 breast cancer cell growth with IC50 of 7 nM, exhibits low hERG activity (IC50=1.13 uM).
SRC-3 inhibitor SI-12 (5–50 nM) can significantly inhibit viability, migration and invasion of breast cancer cells in vitro and repress the growth of breast cancer PDX organoids.
SRC-3 inhibitor SI-12 (10 mg/kg/day) was sufficient to repress growth of xenograft tumors derived from MDA-MB-231 and LM2 cells.
SRC-3 inhibitor SI-12 effectively inhibited progression of breast cancer lung metastasis in spontaneous and experimental metastasis mouse models developed from MDA-MB-231 and LM2 cells respectively.
