SSR128129E is a specific, allosteric, small-molecule multi-FGF receptor (FGFR) inhibitor, binds to the extracellular FGFR domain without affecting orthosteric FGF binding, inhibits FGF-induced signaling linked to FGFR internalization in an allosteric manner.
SSR128129E potently inhibits FGF2-induced endothelial cells (ECs) proliferation and migration with IC50 of 31 nM and 15.2 nM respectively.
SSR128129E inhibits FGFR paralogues in various species across the animal kingdom, including Danio rerio (zebrafish), Drosophila melanogaster (fruit fly), Spodoptera frugiperda , and Bombyx mori (silkworm).
SSR128129E does not inhibit binding of VEGF, VEGF-B, placental growth factor, platelet-derived growth factor (PDGF)-BB, or PDGF-CC.
SSR128129E inhibits tumor growth and metastasis in various models, amplifies the activity of anti-VEGFR2 in refractory tumor models.
