Stiripentol (BCX2600) is an antiepileptic drug that acts directly on GABA(A) receptor as a positive allosteric modulator, also is an lactate dehydrogenase (LDH) inhibitor, inhibits lactate production and decreases glioblastoma invasion and growth. Stiripentol can cross the blood–brain barrier for inhibiting cerebral LDHA/B activity in preclinical therapy studies in mice. Stiripentol (150 mg/kg, i.p.) significantly increased survival in P3 tumor‐bearing mice. Stiripentol induced genetic and metabolic alterations in GBM cells. Stiripentol strongly suppressed drug-resistant epilepsy in animal model.
