Biological Activity
T2M-010 is a potent small molecule binder of Triggering receptor expressed on myeloid cells 2 (TREM2) with MST KD of 0.83 uM, functions as an activator of TREM2 signaling.
T2M-010 (25 uM) led to a robust twofold increase in SYK phosphorylation, a canonical downstream readout of TREM2 activation in HEK–hTREM2/DAP12 cells.
T2M-010 significantly enhanced bead uptake, demonstrating a marked increase in phagocytic activity that paralleled the effect of VG-3927, in BV2 cells.
T2M-010 shows capacity to recapitulate protective microglial programs previously linked to genetic or antibody-mediated TREM2 activation.
