TC14012 is a selective and peptidomimetic CXCR4 antagonist with IC50 of 19.3 nM, also is a specific CXCR7 (ACKR3) agonist with EC50 of 350 nM for recruiting β-arrestin 2 to CXCR7. TC14012 antagonizes the activation, migration, and antiapoptotic responses of CXCL12 in chronic lymphocytic leukemia B cells. TC14012 significantly recruited β-arrestin to CXCR7 in CXCR7-expressing cells and activated extracellular signal-regulated kinase (ERK) in neonatal rat cardiomyocytes.
