TD-198946 is a small molecule disease-modifying osteoarthritis compound, effectively repairs cartilage by promoting chondrogenic differentiation and halt osteoarthritis (OA) progression, exerts its effect through the regulation of Runx1 expression.
TD-198946 strongly induced chondrogenic differentiation without promoting hypertrophy in cell and metatarsal organ cultures.
TD-198946 prevented and repaired degeneration of the articular cartilage.
TD-198946 (>1 nM) dose-dependently enhanced glycosaminoglycan (GAG) production, particularly hyaluronan, whereas chondrocyte differentiation was not impacted.
When combined with transforming growth factor β3 (TGFβ3), TD-198946 promoted chondrocyte differentiation and production of cartilaginous matrices at doses <1 nM as judged by SOX9, S100, and type 2 collagen upregulation.
