TERT inhibitor NU-1 is an irreversible telomerase reverse transcriptase (TERT), targets the non-homologous end joining (NHEJ) DSB repair pathway.
NU-1 decreases cell viability in telomerase-positive human cancer cell lines, including breast cancer MCF7 (LD50=21 uM), impacts DNA damage pathway gene expression.
NU-1 enhances the chemo-sensitivity of telomerase-positive cancer cells.
NU-1 induces radiosensitivity and cellular senescence, induces persistent DNA damage foci and delays double-strand break repair after irradiation in telomerase-positive cells.
NU-1 dramatically enhanced the effects of radiation, leading to immune-dependent tumor elimination in CT26 tumors in BALB/c mice.
