Biological Activity

TH1027 is a highly potent and specific inhibitor of TLR8, dose-dependently inhibited R848-or ssRNA06-induced TLR8 activation with IC50 of 30 nM.
TH1027 binds to human TLR8 (hTLR8) with Kd of 225 nM.
TH1027 inhibits TLR8-mediated signaling without affecting other TLRs including TLR1/2, TLR2/6, TLR3, TLR4, TLR5, TLR7, and TLR9.
TH1027 (1 uM) completely abolished the elevation of TNF-α and IL-8 mRNA levels induced by R848 in HEK-Blue TLR8 cells.
