TLR7 agonist 20 is a potent, selective TLR7 agonist with EC50 of 12 nM and 17 nM for hTLR and mTLR7 in cell-based reporter assay, respectively.
TLR7 agonist 20 displays no significant activity against TLR8 (EC50>5 uM).
TLR7 agonist 20 significantly induces secretion of cytokines IL-6, IL-1β, IL-10, TNFa, IFNa, and IP-10 in human and mouse whole blood. shows a significant secretion of IFNα and TNFα cytokines in pharmacokinetic and pharmacodynamic studies in mice.
TLR7 agonist 20 shows strong synergistic antitumor activity with complete tumor regression in 8/10 mice dosed using the intravenous route, when combined with aPD1.
