TPEN (TPEDA) is a potent, cell-permeable inhibitor of LIN28 that abolishes LIN28-mediated oligouridylation with IC50 of 2.5 uM, also is a specific cell-permeable heavy metal chelator.
TPEN (TPEDA) completely inhibits LIN28-mediated oligouridylation at 40 uM.
TPEN (TPEDA) targets only the C-terminal zinc-knuckle domain (ZKD) domain of LIN28 and causes drastic structural changes of the zinc knuckles and RNA-binding residues in the ZKD.
TPEN (TPEDA) was previously shown to induce apoptosis by chelating Zn2+ in cells.
