TRDMT1i (YW1842, YW-1842) is a potent small molecule inhibitor of RNA methyltransferase TRDMT1, blocks TRDMT1-mediated m5C formation in DNA-RNA hybrids in vitro and reduces the viability of cancer cell lines.
TRDMT1i (YW1842, YW-1842) exhibits -m5C inhibitory activity almost equivalent to TRDMT1 depletion at the concentration of 2.5 uM.
TRDMT1i (YW1842, YW-1842) specifically sensitizes cells to DNA damage agents in a TRDMT1-dependent manner.
TRDMT1i selectively induces synthetic lethality in MutLα-deficient cells, but not MutLβ and MutS-deficient cells.
TRDMT1i (100 or 200 mg/kg, oral gavage) inhibits growth of MLH1-deficient tumors in BALB/c mice bearing CT26 Mlh1-KO tumors.
