TTZ-2 (zDHHC2 inhibitor TTZ2) is a specific enzymatic inhibitor of ZDHHC2, inhibits human zDHHC2-mediated substrate (SNAP25) S-acylation in vitro with IC50 of 1.3 uM.
TTZ-2 inhibits zDHHC2-mediated SNAP25 S-acylation in cells with IC50 of 19 uM in HEK293T cells transfected with plasmids encoding GFP-tagged SNAP25b and HA-tagged zDHHC2.
TTZ-2 shows no effect on S-acylation of SNAP25 by zDHHC3 and zDHHC7.
TTZ-2 (25 uM) significantly inhibits PSD95-mCHERRY S-acylation by zDHHC2 and zDHHC15 in HEK293T cells.
