Tagtociclib hydrate (PF-07104091) is a highly potent CDK2-selective inhibitor with Ki of 1.16 nM, shows potential for the treatment of cyclinE-amplified cancers. PF-07104091 displays 100-fold selectivity against CDK1, 200 to 400-fold selectivity against CDK4 and CDK6, 170-fold selectivity against CDK9, and greater than 500-fold selectivity against GSK3B. PF-07104091 demonstrated a dose-dependent tumor growth reduction with 86 percent maximal effect in ovarian cancer model and demonstrated single-agent efficacy across multiple ovarian models.
