Teloxantron (Telomerase inhibitor TXT4) is a potent small molecule inhibitor of telomerase activity with IC50 of 9.61 uM, 7-fold lower than BIBR1532 (Cat# PC-45170), inhibits the processivity of telomerase with preferential DNA damage on telomeres.
TXT4 induces remarkable and time-dependent induction of DNA damage in TERT-positive (A549 and H460), but not in TERT-negative NHBE normal human bronchial epithelial and ALT-positive U2OS osteosarcoma cell lines.
TXT4 caused an increase in the phosphorylation of ATM and ATR, two main DDR signaling kinases, in both A549 and H460 cells, stimulats apoptotic cell death via the intrinsic and extrinsic pathway in lung cancer cells.
