UBCS385 (Sirt4 inhibitor 60) is a potent, selective inhibitor of mitochondrial deacylase Sirtuin 4 (Sirt4) with IC50 of 0.9 uM.
UBCS385 displays weak effect on Sirt3 and high selectivity against Sirt1, −2, −5, and −6.
UBCS385 inhibits Sirt4 through binding to active site regions overlapping with the acyl-peptide binding regions but not with the NAD+ site.
