Biological Activity
Sebrinoflast (VENT-02) is a potent, selective, orally bioavailable, central nervous system (CNS)-penetrant, and reversible inhibitor of NLRP3, inhibits IL-1β and IL-18 release in ATP stimulated LPS-primed whole blood with IC50 of 0.19 and 0.17 uM respectively.
VENT-02 inhibitsIL-1β and IL-18 release in a concentration-dependent manner following LPS stimulation of human iPSC-derived microglia with IC50 of 6.1 nM.
VENT-02 inhibits IL-18 release in following LPS stimulation of human iPSC-derived microglia with IC50 of 6.3 nM.
VENT-02 doesnot inhibit TNFa release.
VENT-02 binds to a specific pocket on the monomeric form of NLRP3, stabilizing the inactive conformation and preventing the conformational changes necessary for oligomerization and full inflammasome activation.
