VU041 is a selective inhibitor of mosquito Anopheles (An.) gambiae and Aedes (Ae.) aegypti inward rectifier potassium (Kir) channels with IC50 of 496 nM for AnKir1. VU041 preferentially inhibited AeKir1 over AeKir2B in two-electrode voltage clamp experiments in Xenopus oocytes. VU041 does not inhibit mammalian Kir1.1, Kir4.1, Kir6.2/SUR1, and Kir7.1 with IC50 of <30 uM. VU041 is equally toxic to insecticide-susceptible and -resistant strains of mosquitoes. VU041 inhibits renal excretory function in mosquitoes, overcomes insecticide-resistance.
