Biological Activity

XL-3156 is a potent, selective, cross-species, allosteric inhibitor of cyclic GMP-AMP synthase (cGAS), targets both the orthosteric and allosteric sites, effectively inhibits cGAS-DNA phase separation.
XL-3156 efficiently reduces the h-cGASCD-DNA interactions (EC50=783 nM) as well as phase separation in electrophoretic mobility shift assay (EMSA).
XL-3156 induces a morphological shift of condensates from LSPS to LLPS by reducing the degree of phase separation.
