YK-4-279 is a specific small molecule inhibitor of EWS-FLI1 interaction with RNA helicase A with SPR KD of 9.48 uM for EWS-FLI1, reduces EWS-FLI1 functional activity, also reduces EWS-FLI1 modulated transcription. YK-4-279 specifically inhibits Ewing's Sarcoma Family Tumors (ESFT) cell growth (IC50 values between 0.5 to 2 μM) and induces apoptosis, but not cell death in the non-transformed cells. YK-4-279 (60-75 mg/kg) inhibits ESFT xenograft growth in CHP-100 xenograft tumors.
