YNT-185 is a potent, selective orexin type-2 receptor (OX2R) agonist with EC50 of 28 nM, 100-fold selectivity over OX1R.
YNT-185 epolarized OX2-expressing histaminergic neurons in mouse brain slices.
YNT-185 suppressed cataplexy-like episodes in orexin knockout and orexin neuron-ablated mice, but not in orexin receptor-deficient mice.
Peripherally administered YNT-185 also promotes wakefulness without affecting body temperature in wild-type mice.
