Biological Activity
YS-01 is a potent, specific small molecule inhibitor of Cl-/HCO3- exchanger Pendrin (SLC26A4), potently inhibits pendrin-mediated Cl-/SCN-, Cl-/I-, Cl-/HCO3-, and Cl-/OH- exchange activity in a dose-dependent manner.
YS-01 showsno cytotoxicity up to 30 μM in NIH3T3 and CHO-K1 cells and more potently inhibits pendrin activity than PDSinh-A01 and PDSinh-C01.
YS-01 inhibits both human and mouse pendrin-mediated Cl−/I− exchange with nearly identical potency.
YS-01 weakly inhibits Cl−/HCO3− exchange activities of SCL26A3 and SLC26A6 with IC50 greater than 100 μM and does not affect SCL26A7 and SLC26A9.
YS-01 significantly attenuated OVA-induced airway hyperresponsiveness and reduced the numbers of eosinophils and neutrophils in the bronchoalveolar lavage fluid of OVA-sensitized mice.
