zDHHC2 inhibitor TTZ1 (Tetrazole-1, TTZ-1) is a specific enzymatic inhibitor of palmitoyltransferase ZDHHC2, inhibits human zDHHC2-mediated substrate (SNAP25) S-acylation in vitro with IC50 of 4.7 uM.
TTZ-1 inhibits zDHHC2-mediated SNAP25 S-acylation in cells with IC50 of 15 uM in HEK293T cells transfected with plasmids encoding GFP-tagged SNAP25b and HA-tagged zDHHC2.
TTZ-1 shows no effect on S-acylation of SNAP25 by zDHHC3 and zDHHC7.
TTZ-1 (25 uM) significantly inhibits PSD95-mCHERRY S-acylation by zDHHC2 and zDHHC15 in HEK293T cells.
TTZ-1 restores ACSL4 protein, reactivates ferroptosis, and reverses enzalutamide resistance in CRPC cell lines and patient-derived xenograft models.
