Biological Activity
ZQ105 is a potent, highly selective complement receptor C5aR2 agonist with KD of 1.25 uM, does not activate C5aR1, induces β-arrestin1 recruitment to C5aR2 with pEC50 of 6.02.
ZQ105 showsno activity toward C3aR, a complement receptor activated by C3a.
ZQ105 increases intracellular reactive oxygen species (ROS) production, upregulated CD11b expression, and enhanced LPS-induced IL-6 production in human neutrophils.
ZQ105 selectively enhances surface expression of CD10 and LPS-induced IL-1β production in neutrophils.
ZQ105 induces significant reduction in surface expression of C5aR2 in human neutrophils.
ZQ105 induces marked perinuclear redistribution and increased fluorescence intensity of the endosomal and lysosomal markers EEA1, RAB7, and LAMP1, indicating that C5aR2 activation modulates endo-lysosomal organization.
