MI-181 (Mitotic inhibitor-181) is a potent M-phase inhibitor with mitotic arrest IC50 of 23 nM and cell death IC50 of 17 nM, inhibits tubulin polymerization and modulates cilia length and biogenesis.
MI-181 is a potent cell cycle-specific inhibitor, which arrested cells in mitosis, activated the SAC, and induced an apoptotic cell death with faster kinetics than taxol.
MI-181 is active in a broad array of cancers, especially melanomas (BRAFV600E melanomas, IC50=18-90 nM).
MI-181 is effective at rescuing cilia length and ciliation defects in cells that had been treated with the intraflagellar transport inhibitor Ciliobrevin D or the O-GlcNAc transferase inhibitor OSMI-1.
MI-181 induces an increase in cilia length and restores ciliation in cells with compromised shortened cilia at low nanomolar concentrations.
MI-181 restores ciliation and cilia length in cells with a compromised IFT system.
