MNS1-Leu is a novel pyrazole-based small molecular inhibitor of STAT3 pathway, potently inhibit IL-6-induced STAT3 phosphorylation at 10 μM in dBT114 cells.
MNS1-Leu shows selective toxicity towards adult GBM and pediatric high grade glioma cells with IC50 values of 2.99 μM for MC-BT114 cells, 3.83 μM for MC-PED8 cells and 2.42 μM for SF7761.
MNS1-Leu (10 uM) effectively inhibits STAT3 activation and nuclear translocation in both adult GBM and pediatric HGG cells.
MNS1-Leu inhibits STAT3 activation via IL-6-mediated STAT3 pathway and not direct JAK2.
MNS1-Leu (0.5-2.5 uM) induces apoptosis and inhibit migration in adult GBM cells.
MNS1-Leu (10 mg/kg) slows pHGG growth in vivo and reduces STAT3 activation in pediatric HGG PDX model, with no toxicity and good stability in human plasma.
