WP1066 is a potent, cell-active STAT3 pathway inhibitor, inhibits human malignant glioma U87-MG and U373-MG cells in vitro with IC50 of 5.6 and 3.7 uM.
WP1066 activated Bax, suppressed the expression of c-myc, Bcl-XL and Mcl-1, and induced apoptosis.
WP1066 inhibited the growth of subcutaneous malignant glioma xenografts during the 30-day follow-up period via systemic intraperitoneal administration.
WP1066 inhibited the phosphorylation of JAK2 and also degraded JAK2 protein in AML cells, thereby blocking its downstream STAT and PI3K pathways.
