ARN16186 is a potent, specific small molecule inhibitor of N-Acylethanolamine-hydrolyzing acid amidase (NAAA) with IC50 of 23 nM.
ARN16186 displays little to no activity against human acid ceramidase (h-AC) and fatty acid amide hydrolase (h-FAAH) at 10 uM.
ARN16186 inhibits NAAA in a noncovalent manner.
ARN16186 increases PEA and OEA levels and reduces NF-kB and STAT3 DNA-binding activity in the spinal cord of mice.
ARN16186 reduces mononuclear cell infiltrate in the spinal cord and affects the CD4+T lymphocyte population, reduces the progression and severity of symptoms in EAE mice model.
