DSR-6434 (DSR6434) is a highly potent, specific TLR7 agonist with EC50 of 7.2 nM (hTLR7) and 4.6 nM (mTLR7) for stimulating reporter gene activity in HEK293 cells expressing TLR7.
DSR-6434 shows no acitvity in HEK293 cells expressing the structurally similar hTLR8 or hTLR9.
DSR-6434 induces type 1 interferon and activation of T and B lymphocytes, NK and NKT cells.
DSR-6434 intravenous administration enhances the efficacy of ionizing radiation (IR) and leads to improved survival in mice bearing either CT26 or KHT tumors.
DSR-6434 in combination with IR primes an antitumor CD8+ T-cell response leading to improved survival in syngeneic models of colorectal carcinoma and fibrosarcoma.
