ELOVL1-IN-27 is a highly potent, selective, CNS-penetrant inhibitor of elongation of very long chain fatty acid 1 enzyme (ELOVL1) with IC50 of 13 nM (C26:0 VLCFA inhibition in HEK cell).
ELOVL1-IN-27 selectively inhibits ELOVL1, reducing C26:0 VLCFA synthesis in ALD patient fibroblasts, lymphocytes, and microglia.
ELOVL1-IN-27 reduced C26:0 very long chain fatty acid (VLCFA) concentrations to near-wild-type levels in blood and up to 65% in the brain, a disease-relevant tissue in mouse models of adrenoleukodystrophy (ALD).
